چکیده

This paper reports the use of affinity capillary electrophoresis (ACE) in the measurement of binding constants involving low molecular weight receptors, using vancomycin as an example.2'3 In this system, it was straightforward to measure binding constants of four compounds (two pairs of enantiomeric peptides) to vancomycin simultaneously, and the technique can, in principle, be extended to larger numbers of ligands. We used vancomycin obtained from Streptomyces orientalis and N-acyl-D(r,)-alanyl-o(l)-alanines as a model system.a This system D'D r t €

Using Affinity Capillary Electrophoresis To ldentify the Peptide in a Peptide Library that Binds Most Tightly to vancomycinr

This paper describes a procedure, based on competitive binding, for identifying tight-binding ligand(l)Ig a receptor in mixtures of equimolar ligand candidates using affinity capillary electrophoresis (ACE). Vancomycin and a small library of 32 peptides were used as a model syatem to illushate the procedureThis procedure should be applicable to peptide mixtures of greater complerity than the on...

Kinetic CE: foundation for homogeneous kinetic affinity methods.

Kinetic capillary electrophoresis (KCE) is defined as capillary electrophoresis of species that interact during electrophoresis. KCE can serve as a conceptual platform for development of homogeneous kinetic affinity methods for affinity measurements (measurements of binding parameters and quantitative measurements) and affinity purification (purification of known molecules and search of unknown...

Multiple-injection affinity capillary electrophoresis to examine binding constants between glycopeptide antibiotics and peptides.

Multiple-injection affinity capillary electrophoresis (MIACE) was used to determine binding constants (K(b)) between vancomycin, ristocetin, and teicoplanin from Streptomyces orientalis, Nocardia lurida, and Actinoplanes teichomyceticus, respectively, and fluorenylmethoxycarbonyl (Fmoc)-(Gly, Ala, Val, and Phe)-D-Ala-D-Ala peptides. In this technique, separate plugs of sample containing non-int...

Partial-filling affinity capillary electrophoresis techniques to probe the binding of glycopeptide antibiotics to D-Ala-D-Ala terminus peptides.

This work is an overview of our use of affinity capillary electrophoresis (ACE) to estimate binding constants between D-Ala-D-Ala terminus peptides and the glycopeptides vancomycin (Van) from Streptomyces orientalis, teicoplanin (Teic) from Actinoplanes teicomyceticus, and ristocetin A (Rist) from Nocardia lurida. In these studies, modifications in the ACE technique, including partial-filling A...

Recent Developments in Affinity Capillary Electrophoresis: A Review

Affinity capillary electrophoresis (ACE) is a versatile analytical technique that has been shown to be an efficient and accurate tool to probe non-covalent interactions and to determine binding and dissociation constants between receptors and ligands. ACE uses as its basis the change in migration time of a receptor upon binding to a ligand generally found in the electrophoresis buffer. Subseque...
